Her ex-boyfriend, who was killed in a vehicle crash on Monday a day after Dr Preethi Reddy disappeared, is a suspect.

She was last seen at Macdonalds following the Mardi Gras parade, and last spoke to her family at around 11am on Sunday, letting them know she was heading home.

"Further inquiries have established that Preethi (Reddy) stayed at a hotel on Market Street, Sydney CBD, on Sunday with a man known to her", the police said.

Hurstville Dental & Physio Centre manager Dave Vagadia told news.com.au that Dr Reddy worked at the surgery as a dentist for about five months until March 2018.

In a shocking twist, Narde died on Monday night - a day after Dr Reddy was reported missing - in a head on collision with a truck near Tamworth in what police believe was a deliberate act.

The man is believed to be her former boyfriend who was also attending the dental conference, a police source told AAP.

"I can not qualify exactly what has happened and I won't put it out there because if we're wrong and it's not substantiated then we're doing a disservice to Preethi and her family", Detective Superintendent Gavin Dengate told ABC News.

Police officers searched the auto after it was found at Strachan Lane.

Preeti went to St. Leonards for a dental conference where she had met Narde, who was her colleague and friend earlier.

Her family didn't know if she attended the parade, where she stayed on Saturday night, where she was on Sunday morning, or what happened to her vehicle.

Preethi Reddy was last seen waiting in line at McDonald's in George Street on Sunday.

The New South Wales police said investigation was in progress and details should be verified before they were shared.

Her family reported that Harsh has been harassing her from some time.

SMH reports that police confirmed Reddy's ex-boyfriend has died following a vehicle accident near Tamworth, shortly after speaking with detectives regarding Reddy's disappearance on Sunday.

"The freakish thing I noticed that night was that he took down his Facebook shortly after the conference".

Preethi was captured on CCTV inside McDonald's at the Strand Arcade in central Sydney about 2.15 a.m. on Sunday and appeared to leave alone.

"It's very important for us because, right now, we are working for Preethi and we are working for Preethi's family as well", Detective Superintendent Gavin Dengate said on Wednesday.

"Prior to this unthinkable event, she had been the happiest I'd ever seen her - she was living her best life", she added.

"It is highly out of character for Preethi to not contact family and loved ones", one post said.
